Abstract

Rahman MM, A Pangastuti, R Rakhmawati. 2011. Mutagenic test of chloroform extract partition result of ambre leaves (Geranium radula) to Salmonella typhimurium TA98, TA100 and TA 1535 and profile of the most active part of chemical compound. Biofarmasi 12: 1-10. The purpose of the research was to know the mutagenic effect of partition chloroform extract of Ambre leaves to the tested bacteria Salmonella typhimurium TA98, TA100 and TA 1535 and profile of the most active part of bioactive showing the biggest mutagenic characteristic. The research used Ames method with mutated bacteria S. typhimurium TA98 as frameshift mutation indicator and S. typhimurium 100 and 1535 as substitution mutation of base pair indicator. Tested sample was the result of partition chloroform extract of Ambre leaves of the dissolved and non dissolved part in wash-benzene at concentration of 10, 100, 500, and 1000 μg/ml. Mutagenic test result showed chloroform extract of Ambre leaves of the dissolved part in wash-benzene used S. typhimurium TA100 and 1535 could cause substitution mutation of base pair in concentration 100 μg/mL and 500 μg/mL and used S. typhimurium TA98 could cause frameshift mutation in concentration 10 μg/mL. Mutagenic test result showed chloroform extract of Ambre leaves of non dissolved part in wash-benzene used S. typhimurium TA100 and 1535 couldn’t cause substitution mutation of base pair and used S. typhimurium TA98 could cause frameshift mutation in concentration 10 μg/mL. Result of detection on result compound of partition chloroform extract of Ambre leaves of dissolved part in wash-benzene on KLT plat with various detection of sprayed compound showed a positive result for detection of terpinoid used vanillin-H2SO4 and flavonoid used FeCl3. Terpenoid and flavonoid presumed as compound that causing mutation.
