The objective of this research was to get information about the characteristics of mutant seed from Hoya diversifolia and to obtain the best media for its germination. The research was conducted at Treub Laboratory, Center for Plant Conservation Bogor Botanic Gardens LIPI, Bogor. The research was arranged in a Completely Randomized Design with two treatment factors and three replications. The first factor was varieties that consisted of a mutant and parents, while the second factor was germination media that consisted of sand, cocopeat, and a mixture media between sand and cocopeat (1:1). The results showed that the highest seed germination of mutant from H. diversifolia on a mixture media between sand and cocopeat (1:1) for two weeks after planting was 83.33%. The height of initial seedling one week after planting was influenced by varieties. The mutant of H. diversifolia had a higher seedling height than its mother stock. However, two weeks after planting, the seedling height was influenced by the media. Cocopeat or a mixture media between sand and cocopeat (1:1) were the best media for producing the highest seedling of mutant and its mother from H. diversifolia.
